Biology of Germination: What Happens Inside the Seed
To understand why certain methods fail, you need to understand what happens inside the seed before anything is visible from outside. The cannabis seed stores its energy primarily in the cotyledons, not in the endosperm like cereals. When hydrated, specific enzymes trigger a biochemical cascade that converts those starches into sugars available to the embryo.
Imbibition (0–12 h)
The seed absorbs water through the seed coat. Hydrophilic proteins and cell walls swell visibly. Metabolism begins to reactivate slowly.
Enzymatic reactivation (12–48 h)
Gibberellins activate amylases that break starches into sugars. Proteases convert proteins into amino acids. Oil bodies release fatty acids as additional energy reserves.
Root emergence (24–72 h)
The radicle — the first root — breaks the seed coat and emerges. This is germination in the technical sense. The root grows downward by gravitropism.
Cotyledon deployment (48–120 h)
The hypocotyl pushes the cotyledons upward forming a protective hook. On detecting minimal resistance, it straightens and the cotyledons unfold, beginning photosynthesis.
Cannabis performs epigeal germination: the hypocotyl elongates and carries the cotyledons above ground level, like beans — unlike peas (hypogeal germination). This explains why exact sowing depth matters: too deep and the hook can't reach the surface.

The most recommended method by the grow community. Allows you to see exactly when and how each seed germinates. Requires checking every 12–24 hours.
- Moisten 2 kitchen paper towels (unprinted) with pH 6.0–6.5 water. Wring out: moist but not dripping.
- Place seeds spaced on one towel, cover with the other and put on a plate.
- Seal in a resealable bag or container with a lid. Leave a little ventilation.
- Store in a warm, dark place at 21–25°C.
- Check every 12–24 hours without touching the roots.
- Transplant when the radicle reaches 0.5–1.25 cm.
Pure cotton wool entangles roots in its fibres. Use smooth kitchen paper. Heavily textured paper can do the same if the root grows into the gaps.
The simplest method. Water softens the seed coat and accelerates imbibition. Critical limit: maximum 24–32 hours. Beyond that, oxygen deficit in the water can drown the embryo.
- Fill a glass with warm water at 22°C, pH 6.0–6.5.
- Add seeds and place in a warm, dark spot.
- Wait for the radicle to appear (24–48 hours).
- Once you see the root (0.5 cm), transfer immediately to substrate.
Mimics natural germination. Eliminates the risk of damaging the radicle during transplant, but offers no visual control. Exact depth: 1 cm (rule: 2× the seed diameter).
- Prepare light, porous substrate at pH 6.0–7.0, low in nutrients.
- Make a hole exactly 1 cm deep with a pencil or similar.
- Place seed, cover lightly without compacting.
- Maintain constant moisture: moist but never waterlogged.
- Wait 3–10 days. No activity after 10 days: seed likely not viable.
Ideal for transitioning to hydroponics or coco. Rockwool is naturally alkaline (pH 7.5–8): pre-soak with pH 5.5–6.0 water before use. Jiffy pellets are simpler but have limited root space.
- Pre-soak the rockwool cube with pH 5.5 water. Drain excess.
- Insert seed into the pre-formed hole at 1 cm depth.
- Keep at 22–26°C with 70–80% RH. Ventilate daily.
- Transplant when the root protrudes from the cube.
Verified Optimal Parameters
A 2022 study on Cannabis sativa (MDPI, Hemp) calculated cardinal germination temperatures: biological minimum 3.4°C, calculated optimum 29.6°C, biological maximum 42.6°C. The practical optimum of 20–25°C is more conservative because at 29°C the risk of rapid desiccation and fungal contamination increases significantly outside laboratory conditions.
Thermal stability matters more than the exact value: abrupt day/night fluctuations are more damaging than a stable temperature slightly outside the optimum.

Cannabis is photoblastically neutral: it can germinate in darkness, light or intermediate illumination. The universal recommendation of darkness is not a biological requirement but for thermal stability and mimicking the natural underground germination environment.
The 7 Mistakes That Kill Seeds
Waterlogged substrate = oxygen blockage. Dripping paper = mould. The medium must be moist, never flooded. This is the #1 mistake according to the grow community.
Below 18°C: slow enzymes, process stalled. Above 30°C: heat stress. Day/night fluctuations more damaging than a stable imperfect temperature.
The radicle is extremely delicate. Never touch with fingers. Always use tweezers. The plant will shed the shell itself: do not intervene unless absolutely necessary.
In substrate: exactly 1 cm. Deeper and the hypocotyl hook can't reach the surface. Shallower and the seed is exposed.
Flowering substrate: toxic for seeds due to excess nutrients. Garden soil: unknown composition. Use light peat or a specific germination mix.
Water doesn't contain enough oxygen for the embryo after 32 hours. Without oxygen, the embryo drowns. Remove when you see the root or at 24 hours.
Pure cotton fibres trap the radicle as it grows. Use smooth kitchen paper, not pharmacy cotton wool.
Signs of Success and When to Transplant
Technical germination occurs when the radicle breaks through the seed coat. But our action point is when the root is long enough to handle without risk: between 0.5 and 1.25 cm.
Transplanting too early (root 2–3 mm) is risky because the root is still fragile. Transplanting too late (root 2+ cm in paper towel) increases the risk of the root getting trapped or growing in the wrong direction.

Prepare moist substrate, 1.25 cm hole. Use tweezers. Place the root pointing down. Cover lightly without compacting. Temperature 22–26°C, 65–75% RH, soft lighting for the first hours.

The freezer (-18°C) can extend long-term viability, but only if seeds have less than 8% internal moisture. With residual moisture, ice crystals destroy embryo cells. When removing from the freezer: let seeds reach room temperature before opening the container to prevent condensation on the seeds.
